Grapefruit drug interactions: How does that work again?

KEY POINT

Numerous drugs have been demonstrated to be affected by the concomitant administration of grapefruit juice since the relationship was first recognized in 1989. It appears that other mechanisms (beyond the well-recognized cytochrome P450 [CYP] 3A4 isoenzyme inhibition) are involved in at least some grapefruit-related drug interactions and that at least some of these grapefruit interactions extend to other fruits.
